Level 16 is available after completing Level 15 in Gemcraft Chapter 0 (Gem of Eternity). Although there are 6 paths the monsters can come from, they will all come from the infested tower. This is also the first arcane guardian level. This level has 26 waves. The gem types available in this level are poison, triple damage, splash, slow, reduce armor, and shock. A possible strategy is to have dual gem mastery on level 1-3 and use any grade 4 dual gem to supergem. Combine the grade 4 with grade 1s of the same colour.

Before you kill the last monster, you must destory both arcane locks, which can be destroyed by dropping a grade three or higher gem bomb on it. After all the levels are completed, the Arcane Guardian comes out. It starts out with a massive amount of armor level, but every time you destroy one of the Arcane Minions, the armor level decreases until it reaches zero. This Arcane Guardian is immune to slowness.
- 1 Arcane Guardian (30000 HP, 200 Armor, 0.3 Speed, 2000 Mana to banish 1st time, +50% banishment cost every lap) -Immune to slow
- 12 Arcane Minions (800 HP, 10 Armor, 0.4 Speed, 600 Mana to banish)
After you complete Level 16, your path continues north towards Level 17 .
